Hemolysis and arginase in an in vitro model of thrombotic microangiopathy. Blood cells that were preincubated with Shiga toxin 2 and lipopolysaccharide from E. coli O157:H7 or PBS and suspended in ADAMTS13-deficient plasma were perfused over glomerular endothelial cells. A Hemolysis (OD405 nm) in samples preincubated with Shiga toxin 2 and O157LPS (n = 10) or PBS (n = 10). B Arginase 1 levels in samples preincubated with Shiga toxin 2 and O157LPS compared with PBS control samples C Arginase activity in samples preincubated with Shiga toxin 2 and O157LPS compared with PBS control samples D Correlation between arginase 1 levels and hemolysis in samples preincubated with Shiga toxin 2 and O157LPS (n = 10). Comparisons were performed using Wilcoxon signed rank test (panels A-C) or simple linear regression (panel D). ADAMTS13 A Disintegrin and Metalloproteinase with a ThromboSpondin type 1 motif, member 13, O157LPS Lipopolysaccharide from E. coli O157:H7, PBS phosphate-buffered saline, Stx2 Shiga toxin 2
