From: Molecular target based combinational therapeutic approaches in thyroid cancer
DIM down-regulates estrogen enhanced VEGF expression of HUVECs. ML-1 thyroid cancer cells were seeded at a density of 5×105 cells per well in six-well culture dishes and allowed to adhere overnight. Thyroid cancer cells were then switched to serum free medium and incubated with ± E2 ± DIM or left untreated for 24 hours. The conditioned medium was harvested and HUVECs were cultured in presence of this conditioned medium for 24 hours, followed by whole cell lysate formation. Subsequently, Western blot analysis was performed using anti-VEGF antibody to analyze VEGF expression in HUVECs.
