Characterization of SmyleDCpp65 potency by T cell stimulation in vitro. A 16 h IFN-γ catch assay based on flow cytometry analysis was used to evaluate whether SmyleDCpp65 produced under GMP-like compliant conditions (harvested on day 7 after thawing) could activate autologous CD3+ T cells (n = 3). No antigen (no stimulation) and stimulation with a pp65 peptide mix were used as controls. a Schematic representation of T cell stimulation assays performed with SmyleDC or SmyleDCpp65 (research grade, RG) or with cognate SmyleDCpp65 (GMP1, GMP2, GMP3). b Representative example showing gating and analyses of CD8+ and CD4+ T cells producing IFN-γ after different stimulations. c Bar graph showing the frequencies of CD8+ T cells and d CD4+ T cells producing IFN-γ. Results are indicative of three independent experiments. Bars indicate mean and error bars indicate mean ± SEM. **p < 0.05.
