EGTA inhibited cell adhesion to immobilized CD63 mAb. Top panel: plates were coated with 0.1 ug/ml of the indicated mAb and cell adhesion assays were carried out in the absence (hatched bars) or presence (solid bars) of 10 mM EGTA for 30 min as described in the Materials and Methods. CD63 mAbs: AHN-16, AHN-16.1, AHN-16.5; unclustered control mAb: U131, anti-MCSP mAb: 9.2.27. Bottom panel; plates were coated with 0.1 ug/ml of the CD63 mAb AHN-16 as described in the Materials and Methods, and cell adhesion assays were performed in either 10 mM EGTA, 10 mM EGTA plus 5 mM Ca2+, or 10 mM EGTA plus 10 mM Ca2+ for 30 min as indicated. Results are shown as percent adhesion +/- SEM. A duplicate experiment gave similar results.
