Inhibition of NF-κB activation by H 2 S in M.F. infected U937 cells. U937 cells were pre-treated with the two H2S donors NaHS (1 mM) and GYY4137 (0.1 mM) for 24 h. The cells were then infected with M.F. 18 h following infection the cells were collected and the cytoplasmic fraction was separated from the nuclear fraction. The percentages of NF-kB nuclear subunits p65 (A), p52 (B) and p50 (C) were calculated over the total of NF-κB subunits (nuclear and cytoplasmic) detected as Absorbance (OD) reading at 450 nm. Bars denote the standard deviation. The p-values were calculated as unequal variance t-test of Mycoplasma infected cells relative to 243 media control (*p ≤ 0.05; **p ≤ 0.005) and as unequal variance t-test of Mycoplasma infected cells treated with H2S donors relative to Mycoplasma infected cells (#p ≤ 0.05; ##p ≤ 0.005; ns not significant). The histograms shown are representative of data from three different experiments.
