MTX-NPs increased the number of regulatory B cells in vivo. Beginning 3 weeks after the first immunization with type II collagen (CII), mice were injected subcutaneously with vehicle, free MTX, or MTX-NPs twice per week for 7 weeks (n = 3/group). At 70 days after the first immunization, ex vivo splenocytes were isolated and the number of CD19 + B220 + GL-7 + FAS + and CD19 + CD25 + Foxp3 + cells determined by flow cytometry.
