PNA-A15 suppresses the expression of CSC-like phenotypes. a Cell viability was assessed using a WST-1 assay, and relative cell viabilities were compared to those of untreated controls. b 3D single CSC-enriched populations derived from secondary spheroids of H292 cells were treated with PNA-A15 or scramble 5 μM for 7 days. Spheroid images were captured by microscopy (10 ×) on days 0, 3, and 7 of treatment. Apoptotic and necrotic cell death was assessed by Hoechst 33342/PI costaining. Cells were visualized using fluorescence microscopy (10 ×). c Parental H292 cells were treated with PNA-A15 or scramble 5 μM for 48 h, and the expression levels of CD133, CD44, ABCG2 and ALDH1A1 were assessed by Western blotting. d Enriched CSC H292 cells were treated with PNA-A15 or scrambled 5 μM for 48 h days, and the expression levels of CD133, CD44, ABCG2 and ALDH1A1 were determined by Western blotting.
