17AAG enhanced upregulation of HSP70 in response to sevoflurane injury in vivo and in vitro. Total protein was extracted from rat hippocampal tissue or cultured H4 cells that were treated as in Figs. 1 and 2, respectively. The expression of HSP70 protein was analyzed by Western blot (a and b, rat hippocampus; d and e, H4 cells), with β-actin as loading control. The expression of c human HSPA1A (human) and f rat Hspa1a mRNA was measured by RT-qPCR. n = 3.
