ANP fails to inhibit acute lung injury in GC-A knockout mice. Guanylyl cyclase-A wild-type (GC-A+/+) and knockout mice (GC-A−/−) mice were given an intravenous infusion of ANP or saline alone starting 30 min prior to oral pharyngeal aspiration of saline or psuedomonas 103 (PA103). Animals were sacrificed and lung weight to dry weight ratio was measured 4 h later. PA103 cause a marked increase in pulmonary edema as assessed by lung wet to dry weight in both genotypes. This effect was completely blocked by ANP infusion in wild-type mice, but ANP had no effect in GC-A knockout mice. Data are mean ± standard deviation, n = 4–6 animals per group.
