CD14+ bead-selected monocytes cultured in FBS-supplemented medium represent an optimal clinical-grade compatible approach for DC-10 generation. Adherent fraction of PBMC (empty square) or CD14+ selected monocytes (filled square) were cultured in FBS- or HS-supplemented medium for 7 days to generate tolerogenic Tr1-cell-inducing DC-10. DC-10 yield is shown, measured as: 100× [no. of generated DC-10 cells/no. of plated cells]. One square represents one experiment, lines represent mean value of each data set ± SD. Coefficient of variation (CV) is shown for each data set
