Chemical chaperone inhibited ER stress induced by anti-dsDNA antibodies in HMCs. a, b HMCs were stimulated with anti-dsDNA antibodies. The expression of ATF4 and nuclear translocation of NF-κB p65 were measure by western blot. c, d 4-PBA significantly inhibited the expression of ATF4 and the nuclear translocation of NF-κB p65 induced by anti-dsDNA antibodies. eg 4-PBA significantly inhibited the expression of IL-1β, MCP-1 and TNF-α mRNA as measured by qPCR. hj 4-PBA significantly inhibited the secretion of IL-1β, MCP-1 and TNF-α in the supernatants as measured by ELISA. All experiments were repeated for three times. Data were expressed as mean ± SD, *P < 0.01, versus control media, #P < 0.01, versus anti-dsDNA antibodies group.
