Effect of LRAST on tumor growth in mice with established tumors. (A) LRAST treatment schema in a therapeutic setting. C57BL/6 mice received a s.c. injection with 106 viable mGC8 tumor cells. Three days later, mice in the LRAST group were treated with cyclophosphamide and were reconstituted with spleen cells 24 h later. The same day (day 0), mice were vaccinated with irradiated mGC8 cells (107) and injected with GM-CSF in IFA. One group received no vaccination (no treatment control). The vaccinations with mGC8 and GM-CSF/IFA were repeated every other week for a total of four vaccinations. Tumor growth curves are shown for the individual mice in (B) the LRAST group (n = 5), (C) the mGC8 GM-CSF/IFA-vaccinated group, without cyclophosphamide and reconstitution (n = 5), and (D) the no treatment control group (n = 5). (E) Mean tumor sizes per group shown in B, C and D are plotted (+/- SEM), n = 5 per group. Cyclophosphamide pretreatment tended to delay tumor growth.
