3a-G1 prevents IL-2 driven expansion of CD4+/Foxp3high regulatory T cells. a) Expanded CD4+/CD25+ T cells among IL-2 treated PBMCs present characteristics of regulatory T cells, e.g. CD127-/low and FoxP3high. 3a-G1 interferes with the expansion of these cells. Markers analysed in upper quadrants are obtained after gating on lived cells based on Forward/Side scatter signal. Insert of CD25 staining is gated on FoxP3+ cells overlaid with isotypic control antibody staining. CD25/CD127 quadrant is gated on CD4+ T cells. Percentage of cells from parental gate is indicated in each quadrant. b) Increased ratio of NK:FoxP3high T cells during 3a-G1 driven expansion of human NK cells from PBMCs.
