A) To demonstrate the specificity of TIL-26(GG) for an epitope provided by RCC-26, different tumor cell lines were co-cultivated with the T cell clone in a ratio of 5:1 and supernatants were analyzed 24 h later. B, C and D) HLA-A*0201-positive iDCs were transfected using DMRIE-C with total RNA derived from RCC tumor cell lines. 48 h after transfection and maturation the mDCs were co-cultivated with the TIL-26(GG) T cell clone at a ratio of 5:1 for 24 h. All experiments were carried out in triplicates and activation of T cells was measured by analysis of IFNγ secretion.
