Hirsutanol A induced mitochondrial-independent accumulation of intrinsic ROS in SW620 and MDA-MB-231 cells. Cells were stained with DHE for 1 h after exposure to hirsutanol A for 3 h. The cellular O2 level was monitored by flow cytometry. Results are presented as means ± SD from 3 independent experiments. (** < 0.05 versus *) (A). SW620 and MDA-MB-231 cells were treated with various concentrations of hirsutanol A for 24 h or with 20 μmol/L and 30 μmol/L hirsutanol A for indicated time respectively, and then stained with CM-H2DCF-DA for 1. Results are presented as means ± SD from 3 independent experiments. (** < 0.01 versus *) (B). Raji, HL60, C2, C8 and C6F cells were treated with 20 μmol/L hirsutanol A for 5 h, and then stained with CM-H2DCF-DA for 1 hour. The cellular H2O2 level was monitored by flow cytometry (C).
