The addition of IFN-γ during DC maturation with BCG induces secretion of IL-12 and suppresses IL-10 production IL-10 and IL-12 p70 cytokine were quantified in DC culture supernatants by ELISA after 24 hour maturation with BCG (1.6 × 106 CFU/ml) in the presence or absence of IFN-γ (1000 U/ml). Data shown represent the mean ± SD of IL-10 and IL-12 concentrations from four experiments (A). Increasing concentrations of BCG induces IL-10 secretion by DC in a dose dependent manner (in some donors) that is suppressed by the addition of IFN-γ. Data shown represent the mean ± SD of IL-10 concentrations obtained from three separate but identical experiments (B).
